Immunzellen, die zellulären Bestandteile unseres Immunsystems, bilden das Überwachungssystem nicht nur gegen bakterielle und virale Erreger, sondern auch gegen entartete Krebszellen. Eine Aufgabe der Tumorimmunologie-Forschung ist es, Mechanismen mit denen es Krebszellen gelingt, in direkter oder indirekter Weise das Immunsystem auszuschalten, zu entschlüsseln. Die Chemoimmuntherapie-Arbeitsgruppe beschäftigt sich mit der Erforschung dieser Mechanismen und der Interaktion von Krebszellen und Immunzellen, insbesondere T Zellen. Ein weiterer Schwerpunkt ist die Frage nach den Ursachen der malignen Transformation, sowie die Untersuchung der klonalen Evolution der Tumorzellen. Dabei interessieren wir uns insbesondere für eine Gruppe von körpereigenen Enzymen (APOBECs), die eigentlich eine wichtige Rolle in der Immunabwehr spielen, die aber auch die Erbsubstanz schädigen können und somit direkt zur Tumorentstehung beitragen.

+ Antigen-präsentierende Zellen sind in der Lage, Krebszellen zu internalisieren und Merkmale dieser Zellen an der eigenen Zelloberfläche wie Steckbriefe zu „präsentieren“. Wenn diese Steckbriefe von naiven T Zellen als „fremd“ erkannt werden, reifen diese Zellen zu aktivierten Effektor-Zellen und es werden alle Tumor-Zellen, die diese Steckbrief-Merkmale besitzen, eliminiert. Durch Expression des Moleküls PD-L1 an der Oberfläche der befallenen Zellen und Bindung an den Rezeptor PD-1 werden diese Zellen jedoch vor einer Attacke des Immunsystems geschützt. Eine therapeutische Unterbrechung dieser Bindung, z.B. durch einen Antikörper, macht sie wieder verwundbar gegenüber dem Immunsystem und führt zu einer Reduktion der Krebszellen.

Die zur Forschung eingesetzten Mäuse werden in der Zentralen Tierhaltung (ZTH) der PLUS versorgt.
